Cell wall proteins, such as\u00a0 \u00a0 \u00a0glycosylphosphatidylinositol (GPI)-anchored proteins, are involved in swift and appropriate responses to minor environmental changes in fungi. However, the roles of these proteins in the pathogenic\u00a0<em>S. sclerotiorum<\/em>\u00a0remain largely unexplored. Here, we identified a novel GPI-anchored protein in<em>\u00a0S. sclerotiorum<\/em>, SsGSP1, comprising a Kre9_KNH domain.\u00a0<em>SsGSP1<\/em>\u00a0was upregulated during infection, and the loss-of-function mutants of\u00a0<em>SsGSP1<\/em>\u00a0exhibited the compromised cell wall integrity and reduced \u03b2-glucan content. During inoculation on\u00a0<em>Arabidopsis thaliana<\/em>,\u00a0<em>Nicotiana benthamiana<\/em>, and\u00a0<em>Brassica napus<\/em>, the\u00a0<em>SsGSP1<\/em>-deletion strains demonstrated the decreased virulence. The transgenic<em>\u00a0A<\/em>.<em>\u00a0thaliana<\/em>\u00a0line carrying the sRNA targeting\u00a0<em>SsGSP1\u00a0<\/em>(HIGS-<em>SsGSP1<\/em>) enhanced resistance to\u00a0<em>S. sclerotiorum<\/em>. The\u00a0<em>SsGSP1<\/em>-deficient strains displayed the heightened sensitivity to various stresses, including high osmotic, high oxidative, and heat shock. The yeast two-hybrid and BiFC assays revealed that SsGSP1 interacts with the key stress-related proteins, such as catalase SsCat2, heat shock\u00a0 \u00a0protein Sshsp60, and ABC transporter SsBMR1. Accordingly, transcriptome analysis revealed that the disruption of\u00a0<em>SsGSP1<\/em>\u00a0downregulates the expression of genes involved in oxidative stress response, heat shock response, and chemical agent resistance.
