Two Lys\/Cys residue have rich chemical properties, enabling them to be the key sites in protein post-translational modifications (PTMs), catalytical activities and covalent drugs. They enjoy greatly distinguishing reactivities in different positions. However, many chemical residue-selective modification methods are difficult to label endogenous proteins in live cells, because of biocompatible limitations. In our team, based on the tunable activities of pyridinium-based cationic salts to boost mass ionization, we developed new labeling agents for Lys-\/Cys- selective profiling in different physiological neutral conditions. Then, owing to the positive charge, we discovered and explored a mitochondrial labeling preference in cells. Particularly and recently, we also offered a versatile reaction platform for tracelessly cleavable cysteine-selective peptide\/ protein modification by rationally estimating the electron effect on the nucleophilic halopyridiniums. Altogether, we used pyridinium-based cationic salts to develop new chemical methods to expand the existing toolbox for chemoproteomic methods in drug discovery.<\/p>\n<p><strong>Biography<\/strong><\/p>\n<p>Dr. Rui Wang, is an Associate Researcher at Shenzhen Bay Laboratory (SZBL).
