Treatment options remain limited, and increasing resistance to metronidazole, the current first-line therapy, represents a growing public health concern, while the discovery of new therapeutic candidates has been hindered by limitations in efficient and reliable drug-screening approaches. In this study, an optimized resorufin-based drug screening assay was developed to facilitate the identification and evaluation of compounds with activity against T. vaginalis. The assay incorporates a cell-based plate-reader system, a multidrop dispenser, airtight incubation pods to maintain stable microaerobic conditions, and a fluorescencebased readout of parasite metabolic activity. The method exploits the reductive capacity of T. vaginalis to sequentially reduce resazurin to resorufin and subsequently to dihydroresorufin, producing a measurable fluorescence signal proportional to parasite cell numbers. Several compound series were evaluated using the optimized assay. A series of 7-substituted 7- deazaadenosine analogues demonstrated potent activity against T. vaginalis, with EC50 measurements showing superior inhibitory activity compared with metronidazole, while toxicity toward mammalian cell lines remained low. Subsequent synthesis and structure–activity relationship analysis identified an optimized pharmacophore within this scaffold, and several 7- deazaadenosine analogues rapidly cleared T. vaginalis cultures at concentrations as low as 2×EC50. These findings demonstrate the potential of the optimized resorufin-based assay as an effective platform for anti-Trichomonas drug screening and highlight 7-deazaadenosine analogues as promising candidates for the development of new therapeutic strategies for trichomoniasis.

Biography:

Dr. Amani Alhejely is an Assistant Professor of Immunology, Molecular Parasitology and Microbiology at Jazan University, Saudi Arabia.
